Design Notes

The sequence includes a chloroplast-targeting N-terminal signal peptide from the ribulose bisphosphatase carboxylase/oxygenase activase preprotein from Chlamydomonas reinhardtii and was codon optimized for expression in Chlamydomonas reinhardtii.


Source

The gene sequence was obtained from the genomic sequence of Synechococcus sp. (strain WH8103). The 96 bp sequence for the chloroplast-targeting signal peptide was obtained from in silicon reverse transcription of the MQVTMKSSAVSGQRVGGARVATRSVRRAQLQV sequence of the ribulose bisphosphatase carboxylase/oxygenase activase preprotein from Chlamydomonas reinhardtii and added at the 5' end. This sequence was codon optimized for expression in Chlamydomonas reinhardtii and the entire optimized sequence was finally synthesized by IDT using gBlocks technology.
